چکیده

40 Pulmonary fibrosis is a fatal lung disease characterized by fibrotic foci and inflammatory 41 infiltrates. Short telomeres can impair tissue regeneration and are found both in hereditary and 42 sporadic cases. We show here that telomerase expression using AAV9 vectors shows therapeutic 43 effects in a mouse model of pulmonary fibrosis owing to a low-dose bleomycin insult and short 44 telomeres. AAV9 preferentially targets regenerative alveolar type II cells (ATII). AAV9-Tert-treated 45 mice show improved lung function and lower inflammation and fibrosis at 1-3 weeks after viral 46 treatment, and improvement or disappearance of the fibrosis at 8 weeks after treatment. AAV947 Tert treatment leads to longer telomeres and increased proliferation of ATII cells, as well as lower 48 DNA damage, apoptosis, and senescence. Transcriptome analysis of ATII cells confirms 49 downregulation of fibrosis and inflammation pathways. We provide a proof-of-principle that 50 telomerase activation may represent an effective treatment for pulmonary fibrosis provoked or 51 associated with short telomeres. 52 53

Nos2 deficiency enhances carbon tetrachloride-induced liver injury in aged mice

Objective(s): As a multifunctional molecule, NO has different effects on liver injury. The present work aimed to investigate the effects of Nos2 knockout (KO) on acute liver injury in aged mice treated with carbon tetrachloride (CCl4). Materials and Methods: The acute liver injury model was produced by CCl4 at 10 ml/kg body weight in 24-...

Telomerase activity is required for bleomycin-induced pulmonary fibrosis in mice.

In addition to its well-known expression in the germline and in cells of certain cancers, telomerase activity is induced in lung fibrosis, although its role in this process is unknown. To identify the pathogenetic importance of telomerase in lung fibrosis, we examined the effects of telomerase reverse transcriptase (TERT) deficiency in a murine model of pulmonary injury. TERT-deficient mice sho...

اثر درمانی سیکلوسپورین بر آسیب دیررس ریوی ناشی از گاز خردل در موش

Background and Aim: Among late complications in mustard gas or sulfur mustard (SM) casualties, respiratory problems, especially chronic bronchitis and pulmonary fibrosis are the greatest causes of longterm disability. Cyclosporine has anti-inflammatory effects and is used in other pulmonary fibroses due to other factors. Thus, with respect to the role of oxidative stress in the pathogenesis of...

Mice with Pulmonary Fibrosis Driven by Telomere Dysfunction.

Idiopathic pulmonary fibrosis (IPF) is a degenerative disease of the lungs with an average survival post-diagnosis of 2-3 years. New therapeutic targets and treatments are necessary. Mutations in components of the telomere-maintenance enzyme telomerase or in proteins important for telomere protection are found in both familial and sporadic IPF cases. However, the lack of mouse models that faith...
